Immunohistochemistry

BTA offers an Immunohistochemistry (IHC) and in situ hybridisation (ISH) service for target validation/localisation studies:

High throughput

We use 4 Leica Bond RX autostainers for conventional IHC and multiplex stains for highly reproducible and consistent results

Regulatory compliance

We have experience with conventional IHC for clinical trial outcome measures

Imaging

We use the Leica Aperio AT2 scanner for brightfield whole slide scanning and image hosting on Leica’s Aperio eSlide manager portal.

Image analysis

We can advise on digital image analysis strategies and provide access to image analysis software packages.

 

Multiplex Immunofluoresence

The Akoya PhenoImager™ HT system allows up to 9 colour multiplex immunofluoresence (mIF) and brightfield scanning. We have an existing set of pre-validated panels which require minimum optimisation according to specific tissues of interest. Additionally we can perform panel design and validation according to the needs of individual projects. Additionally, we can give access to Akoya Mantra 2.0 which acts as an optimisation platform for projects aimed for Polaris scanning and can be used to aid groups planning to stain for themselves but scan on the Polaris.

Multi spectral imaging

Imaging on the PhenoImager HT can be applied across a whole slide using the 6-plex, 7 Color Opal Polaris reagent kit or up to 8-plex with deconvoluted region of interest scanning.

High throughput

Staining can be performed fully automated at scale using the Leica Bond RX autostainer platform.

Regulatory compliance

Highly reproducible staining can be applied across studies in line with Good clinical practice (GCP) in the laboratory standards as clinical trial outcome measure.

Hyperplex Immunofluorescence

The Lunaphore COMET is our hyperplex immunofluorescence solution for researchers looking to explore cell-cell biology in greater detail. This system allows the visualisation of up to 40 markers in a tissue sample using the principle of sequential immunofluorescence approach. BTA can provide tissue staining use a range of pre-validated panels with an option to add additional markers of interest.

Digital Spatial profiling

The BTA team is a Certified Service Provider for Nanostring GeoMx Protein and RNA.

The Nanostring GeoMx gives the capability for digital spatial profiling of regions of interest in FFPE sections. These can be profiled for either an 84 plex, 1800 plex or whole transcriptome analysis RNA panels, or a selection of configurable antibody panels made of ~10 plex modules up to ~60 plex.

The Nanostring nCounter can quantify up to 800 RNA targets without amplification from either extracted RNA including FFPE derived material using both off-the-shelf or custom panels available from Nanostring (including CE IVD marked clinical assays).
